Natural abundance 13C nuclear magnetic resonance studies of human plasma high density lipoproteins.

چکیده
High resolution proton-decoupled natural abundance ‘:‘C NMR spectra (at 15.18 MHz) of human plasma high density lipoproteins were recorded at 29”C, Yfi”C, and 43°C. Carbon 5 of unesterified cholesterol and the corresponding carbon of cholesteryl esters yield narrow, well resolved resonances. Values of nuclear Overhauser enhancements and integrated intensities for C-5 of free and esterified cholesterol, C-6 of cholesteryl esters, and the methyl groups on cationic nitrogen of phospholipids were measured. At 35”C, the ratio of integrated intensity of the C-5 resonance of cholesteryl ester to that of cholesterol, 51, is significantly different from the molar ratio of these species, 3:1, determined by chemical analysis. This result requires that at least two pools of cholesterol exist within the HDL particle and suggests that a substantial fraction of the cholesterol is sufficiently motionally restricted so as not to yield a detectable C-.5 resonance. Comparison of integrated intensities of peaks assignable to cholesteryl esters and phospholipids suggests that all or a large fraction of the cholesteryl esters contribute to the high density lipoprotein spectrum. The integrated intensities of peaks reflecting cholesterol and cholesteryl esters do not change over the temperature range 29-43”C, suggesting that no major structural reorganization of these components involving phase changes or formation of highly ordered domains occurs over this temperature range.

متن کاملDetection of human muscle glycogen by natural abundance 13C NMR.
Natural abundance 13C nuclear magnetic resonance spectroscopy was used to detect signals from glycogen in the human gastrocnemius muscle. The reproducibility of the measurement was demonstrated, and the ability to detect dynamic changes was confirmed by measuring a decrease in muscle glycogen levels after exercise and its subsequent repletion. Single frequency gated 1H decoupling was used to ob...
متن کاملLipid dynamics in human low density lipoproteins and human aortic tissue with fibrous plaques. A study by high field 13C NMR spectroscopy.
Natural abundance 13C NMR spectroscopy at 63.4 kG was used to probe the temperature dependence of lipid dynamics in low density lipoproteins (LDL) and in aortic fibrous plaques (i&ma-media sections). Spectra of LDL from normals and a familial type II were essentially indistinguishable. At 37”C, tissue spectra exhibit numerous narrow fatty acid and cholesteryl ester ring resonances, no peaks att...
متن کاملEffects of n-3 fatty acids on the NMR profile of plasma lipoproteins.
The effects of fish oil supplementation (14.5 g n-3 fatty acids/day) on plasma lipoprotein particles in healthy volunteers were assessed by high resolution 13C and 1H nuclear magnetic resonance (NMR) spectroscopy. Resonances not previously observed in the 13C and 1H spectra of plasma and isolated lipoproteins were detected after fish oil ingestion. The 13C resonances, centered at 14.3, 127.1, a...
متن کاملNatural-abundance Carbon-13 Fourier Transform Nmr Studies of Large Molecules
The resolution in proton-decoupled natural-abundance i3 NMRt spectra of large organic and biological molecules is normally considerably greater than in proton NMR spectra, as a result of the large range of '3C chemical shifts 'and the absence of' 3C—' 3C coupling. The Fourier transform NMR technique has partly overcome the problem of very poor sensitivity of natural-abundance l3 NMR. As example...
